Specifications:
The STORM (stochastic optical reconstruction microscope) has a resolution of 20 nm, which is in the size range of biological macromolecules. Better resolution is based on the fact that fluorescent dyes are not excited simultaneously but individually excited and detected in the sample, so that their location can be determined more precisely and the image assembled from the dots is much more detailed. Suitable for studying living cell and fixed samples with unique molecular sensitivity. The microscope uses four laser excitations (405, 488, 561, and 647 nm) so that several different fluorescently labeled molecules can be examined simultaneously. The instrument is also suitable for 2D and 3D imaging. With using TIRF (total internal reflection) illumination, the cell membrane can be examined with particularly high sensitivity. Intended use e.g. examination of the cell surface proteins of tumor cells, lymphocytes, the function of transcription factors that regulate gene function or the organization of chromatin.
Potential use:
- confocal imaging
- dSTORM, 3D STORM imaging, ~20 nm resolution
- fixed and living cell samples with single molecule detection,
- imaging with TIRF illumination
- intracellular trafficking
- large image acquisition
- optical sectioning
